The estrogen injections are 'depot' injections, i.e. you do them once a week into the muscle and the stuff leaks out into the bloodstream gradually, so there is a concentration gradient, i.e. the levels in the blood are highest just after the injection and the lowest just before the next one.
The injection frequency is weekly though, so this 'every third week I feel different' is psychosomatic. It's probably just the euphoria!
