In a Twitter post back in 2023, Riot officially announced that Arcane is officially canon to the League of Legends universe.

No I mean that all the champions are from different points in time fighting without any particular plot in the game. Yeah they are from the same universe obviously, but there is no plot that leads up to them all fighting in this 5v5 arena.
League(game) isn't canon to league (story). League(game) has no story. There is lore and story for the characters that you can play as, as well as the other things in the game, but there is no concept of the game taking place in the wider universe. They don't even attempt to make up an excuse for what's happening in the game.
